1. Metabolism and Cellular Organisation: The True Indicators of Life

The total sum of all chemical reactions occurring within a living organism is known as metabolism. These reactions occur in an interconnected and highly regulated manner, supporting essential life processes such as energy production, biosynthesis, growth, repair and homeostasis. Importantly, no non-living object exhibits metabolism. Stones, metals, water and artificial machines may undergo changes, but none perform coordinated biochemical reactions driven by enzymes and genetic regulation. This makes metabolism a universal and exclusive property of living organisms.

However, advancements in biochemistry have made it possible to perform metabolic reactions outside the body—in what are called cell-free systems. These in vitro systems use isolated enzymes, substrates and cofactors in a test tube or controlled laboratory setup. Under suitable conditions, reactions like glycolysis, DNA replication or protein synthesis can be made to occur outside a living cell. Despite this, such reactions do not make the test tube “alive.” They represent living reactions, but not a living entity.

This distinction is essential. A metabolic reaction occurring outside the body does not possess the hallmarks of life. It cannot grow, reproduce, organise itself, respond to stimuli or maintain homeostasis. It is simply a biochemical event carried out using biological catalysts. Hence, an in vitro reaction cannot be classified as either living or non-living; it is merely a demonstration of metabolic capability extracted from a living system.

What this leads us to is a profound conclusion: metabolism alone cannot define life unless it occurs within the context of a structured, organised cellular system. A test tube full of enzymes and substrates may display metabolic activity, but it lacks the structural and functional organisation of a living organism.

Therefore, the ultimately defining feature of life is cellular organisation. Every living organism, whether unicellular or multicellular, is composed of cells—functional, membrane-bound units capable of coordinating metabolic reactions, regulating internal environment, replicating DNA and generating energy. Cells are the smallest units that exhibit all characteristics of life simultaneously.

In unicellular organisms, the single cell performs all essential functions. In multicellular organisms, specialised cells form tissues, organs and systems that work collectively. Regardless of complexity, life is inseparable from cellular structure. Only within a cell can metabolism be regulated, coordinated and sustained as part of a living system.

Thus, while metabolism is a universal feature of life, the presence of a cellular organisation is what truly distinguishes living beings from all forms of non-living matter, including in vitro biochemical setups. Cells provide the architecture, information system and control network that make life possible.
